Cryptotanshinone, a Potential SIRT3 Inhibitor, Suppresses Colorectal Cancer Proliferation in <i>vitro</i> and in <i>vivo</i>

Cryptotanshinone (CPT) is the main pharmacologically active component of Salvia miltiorrhiza Bunge, and has been demonstrated to have inhibitory effects on a variety cancer cell types. However, target(s) CPT in colorectal (CRC) cells are still be identified. The aim this study was investigate molecular mechanism CPT-induced apoptosis CRC cells. Transcriptome sequencing revealed that 1234 genes were differentially expressed CPT-treated SW480 compared with vehicle control, which associated signaling pathways such as PI3K/AKT, Pathways biological processes include proliferation mitochondrial function. CCK-8, colony formation assay, tumor xenograft models suggested suppressed ability inhibited vitro vivo, while flow cytometry indicated arrested cycle S phase. JC-1, Hoechst Western blot analysis promoted triggered alterations membrane potential. In addition, database showed high SIRT3 expression shorter survival times. Immunofluorescence, blot, docking confirmed able target downregulate protein levels concentration-dependent manner. summary, inhibits vivo.
